Well, as tools per se I use Photoshop CS5 and a regular mouse (I don't have a tablet). Then just play around with the default brushes. The key is modifying the settings, you can get surprising results by messing with the opacity, flow, scatter and so on! To modify the advanced settings (scatter, texture etc.), either press F5 or click on the little folder icon next to the brush icon and size.
It also depends on the look you're aiming for when painting. For a more smooth and blocky look, use round brushes. For a more blurry and "traditional paint" look, use one of the rectangular ones :)
Dunno if I'm the best person to explain painting since I'm not that good at it XD Everything I paint turns out blurry. I tried looking at tutorials on DeviantArt and they helped somewhat, though most seem to be written for those with tablets ;A;

Oh you should try, it's much easier to clean images by vectoring...though it is more time consuming (depends on the image, though). But if you've already traced it wouldn't be too hard for you to pick up vectoring, it's almost the same :)
