If we'll base it from the last Olympics event held in Japan (1998 Winter Olympics in Nagano) as well as the mascots in other countries, it's very likely that Japan will be having an original character as their mascot.
But I have to agree that Doraemon would be the best choice in your list for Olympics mascot. He already has an established international appeal (at least, in many countries other than the US) and, as you said in a previous post, the character definitely represents Japan. I won't even count on Pikachu since the Pokemon creature has been somewhat Westernized, and Hatsune Miku just doesn't appeal to me as a mascot that would actually represent the Olympics.
