• Susumu Kodai: Protagonist of the film. Accepts command of the Yamato after commanding the Earth Cargo Ship Yuki.
• Miyuki Kodai: Daughter of Susumu Kodai and Yuki Mori. Assistant to Sakezo Sado at Dr. Sado's Safari Park.
• Yuki Mori: Wife of Susumu Kodai. Commanding Offcier of the Super-Andromeda Fleet's flagship.
• Shiro Sanada: Head of the Earth Federation Defense Force (originally called "Earth Defense Force").
• Jiro Shima: Shiro Sanada's right-hand man.
• Sakezo Sado: Head of Dr. Sado's Safari Park.
• Analyzer: Sakezo Sado's robot assistant.
• Mi-Kun: Sakezo Sado's cat.
• Kosaku Omura: Executive Officer of the Yamato. Originally Vice-captain of the Cargo Ship Yuki.
• Tasuke Tokugawa: Chief Engineer of the Yamato. Replaces Sho Yamazaki (who served onboard the ship in 2201-2203).
• Minoru Goda: Artillery Specialist of the Yamato. Replacement for Yasuo Nanbu (who served onboard the ship in 2199-2203).
• Atsushi Kobayashi: Leader of the Yamato's Black Tiger squadron (now called the "Cosmo Pulsar squadron"). Also the ship's Chief Helmsman, therefore a replacement for Jiro Shima's brither Daisuke Shima (who served onboard the ship in 2199-2203).
• Miharu Sasaki: The Yamato's Doctor. Replacement for Sakezo Sado (who served onboard the ship in 2199-2203). Also, she's one of the ship's Cosmo Pulsar pilots.
• Ryohei Nakanishi: Chief Communcations Officer of the Yamato. Replacement for Yoshikazu "Gichi" Aihara (who served onboard the ship in 2199-2203).
• Yoichi Sakurai: Main Operator for the wave motion sensor and video system onborad the Yamato. Replacement for Kenjiro Ohta, who served onboard the ship in 2199-2203. Originally served on board the Cargo Ship Yuki.
• Ryo Kamijo: Combat Chief of the Yamato. Originally served on board the flagship Blue Noah during the time the SUS Empire attacked Yuki Mori's ship.
• Saburo Kinoshita: Science Officer of the Yamato.
• Maho Orihara: Head of the team operating the Yamato's Super Brain (formerly known as the ship's "third bridge"). Also, she serves as the ship's Main Bridge Radar Operator, therefore replacing Yuki Mori (who served onboard the ship in 2199-2203).
• Sho and So Tenma: Twin brothers serving as engineers aboard Yamato.
• Juzo Okita: The Original Commanding Officer of the Yamato (Note: This character only appears in flashback moments).

• Metsler: General-Governor of the SUS Empire (also known as the Spacemen United Systems Empire). Commanding officer of the Maya and the SUS Empire's Dreadnaught.
